Aliphatic chain compounds include aliphatic compounds and chain compounds containing other elements or groups. Aliphatic hydrocarbons are hydrocarbons with the basic properties of aliphatic compounds. In aliphatic compounds, carbon atoms are arranged in straight chain, branched chain or cyclic, which are respectively called straight chain aliphatic hydrocarbons, branched chain aliphatic hydrocarbons and alicyclic hydrocarbons. Voltage-gated sodium channels (NaVs) are responsible for action potential initiation and transmission of pain signals. NaV1.8 is specifically expressed in peripheral nociceptors and has been genetically and pharmacologically validated as a human pain target.
LTGO-33 inhibited NaV1.8 in the nM potency range and exhibited over 600-fold selectivity against human NaV1.1-NaV1.7 and NaV1.9. Unlike prior reported NaV1.8 inhibitors that preferentially interacted with an inactivated state via the pore region, LTGO-33 was state-independent with similar potencies against closed and inactivated channels.
